The Embryonic Stem Cell Test
An Efficient in vitro Assay in Developmental- and Reproductive Toxicity Testing
The EST is an ECVAM validated cell-based assay to evaluate embryotoxicity of substances based on cytotoxicity and cardiomyocyte-differentiation endpoints. The latter uses the formation of embryoid bodies (EBs) from mouse embryonic stem cells (ESC) by the hanging-drop (HD) method. The traditional process of EB production and handling technique is labor-intensive and prone to generate non-uniform EBs. In the current webinar a novel hanging-drop platform for efficient and uniform production is presented. Moreover, the predictive performance of the EST is compared to other in vitro embryotoxicity assays such as Whole Embryo Culture (WEC) or Zebrafish. Furthermore complementary endpoints like RNA or Proteomics and refined algorithms for embryotoxic classification in the EST assays are discussed
